She holds an Native American heritage and Lily is a member of the Blackfeet and Aaniiih tribe. Betty and Howard are parents of Lily Gladstone. Her father is of Piegan Blackfeet and Nez Perce heritage, while her mother has an Euorpean descent. She claims relation with the family tree of British Prime Minister William Gladstone from her mother's side. However, she kept the name of her parents private and grew up in Browning town in Montana. Later, her family moved to Seattle city in Washington.

She completed her high school at Mountlake Terrace in Washington. She went to the University of Montana and graduated in 2008 in the field of Fine Arts, majoring in Acting/Directing.

During her graduation years, she took part in Theater of the Oppressed and performed in several productions, such as Riders to the Sea (2006), Richard III (2006), Miss Julie (2007) and Coyote on a Fence (2008).

She acted as Mollie Kyle, who is an Osage who witnessed the assassination of her relatives as a result of her husband’s family’s plans (played by Leonardo DiCaprio). This was in Killers of the Flower Moon – Martin Scorsese’s Modus Operandi, which came out in 2023. Her performance was well regarded, and it was said to be among the film’s high points. Then the remarkable achievement, in 2024, the Golden Globe Award for Best Actress in a Drama Film was won by her and she is the first female Native American to achieve that feat. In that same year, she became the first Native American and only the fourth Indigenous woman ever to be nominated for an Academy Award in the Best Actress category.
